Kept at limited for both. Rationale: Farr 2024 n=60 placebo-controlled RCT missed primary endpoint; the exploratory tertile-stratified finding does not meet the “strong” bar (which would require a confirmatory pre-registered biomarker-stratified trial). The Liu 2025 HNSCC trial is in a different translational frame (immunotherapy adjunct, not aging-rejuvenation), so it does not move the aging-evidence needle. Considered limited-negative per R24 schema since the only RCT primary endpoint was negative — declined because the exploratory subgroup signal is robust enough that “limited-negative” would mis-frame the page (limited-negative is for classes where multiple Phase 2/3 trials reported null/negative for primary endpoints; D+Q has one such trial plus several positive open-label biomarker signals).

Scope: Combined R26c propagation backlog (Martin-Montalvo χ², Cabreiro 2013 dose-specific + live-bacteria numerics) and R25 recency refresh (PubMed + Europe PMC + ClinicalTrials.gov v2 API, 2024–2026 window).
Task A — propagation backlog (R26c) cleared:
- Martin-Montalvo 2013 (10.1038/ncomms3192) verified end-to-end against local PDF: added missing χ²=51.70 for 1% toxic-dose -14.4% mean lifespan, p<0.001 (previously page only carried the χ²=5.46 / 5.83% extension figure for the 0.1% C57BL/6 arm). Added serum metformin concentration (0.45 ± 0.09 mM at 0.1% dose) for in-vivo vs in-vitro dose-extrapolation context.
- Cabreiro 2013 (10.1016/j.cell.2013.02.035) verified against local PDF: added the dose-response (25/50/100 mM → +18/+36/+3% mean lifespan; non-monotonic, peak at 50 mM), the live-bacteria-requirement (axenic culture, UV-killed E. coli, and metformin-resistant OP50-MR each abolish or reverse the life-extending effect; -16% on UV-killed, -37% on OP50-MR), and the folate-disruption mechanism (5-methyl-THF +116%, p=2.5×10⁻⁶ in E. coli) with full citation footnote.
