Ruminococcus bromii is a keystone species for the degradation of resistant starch in the human colon
Design
The investigators compared resistant-starch utilization by four human fecal isolates—Ruminococcus bromii, Bifidobacterium adolescentis, Eubacterium rectale, and Bacteroides thetaiotaomicron—then tested pairwise cocultures on boiled RS2 and retrograded RS3. They also used a fresh fecal sample from one person previously identified as one of two low-RS3 fermenters, alongside three control donors, and added each cultured species separately. This is mechanistic culture/ex-vivo evidence, not a human clinical intervention.
Findings
Ruminococcus bromii and B. adolescentis utilized raw or boiled resistant starch more extensively than E. rectale or B. thetaiotaomicron. In pairwise coculture, however, only R. bromii consistently increased RS2 and especially RS3 utilization by each of the other three species, even in medium that did not support R. bromii growth. It released glucose and maltose from the starch, and its presence stimulated E. rectale growth in coculture.
In the low-fermenting donor’s fecal culture, only addition of R. bromii restored RS3 utilization to the range seen in the three control-donor cultures. The donor sample had undetectable R. bromii-related bacteria and bifidobacteria by qPCR, but also contained oxytetracycline above 20 μg/mL; this likely curtailed growth of all four added strains and makes the single-donor rescue an unusual, confounded ex-vivo test rather than proof of a general human phenotype.
Interpretation
The paper explains how a primary degrader can control a community-level metabolic response and supports a keystone role for R. bromii in RS degradation. It does not show that R. bromii itself is a major butyrate producer, that its abundance predicts gas tolerance, or that supplementation or engraftment improves human health. Pure cultures were tested in triplicate, but the donor rescue rests on one low-fermenting fecal community and requires larger human cohorts. gap/needs-human-replication
